dc.description.abstractPhenolics are a very important but diverse group of compounds found in higher plants. Many phenolic compounds have been identified in the plant kingdom, but the dynamics of their synthesis and the role they play in overall plant development is not well understood. This thesis concerns the separation, identification, quantitation, and physiological studies on the soluble phenolics in young developing pine (Pinus elliottii, Engelm.). High performance liquid chromatography (HPLC) was chosen as a technique for separation and quantitation of soluble phenolic compounds in young plant tissues. The method was developed utilizing phenolic standards and has been shown useful for the separation of several benzoic acids, cinnamic acids, coumarins, and flavonoids. The method utilizes a uBondapak C18 column and the solvents water-acetonitrile-acetic acid, (88:10:2) for coumarins and benzoic and cinnamic acids, and (68:30:2) for flavonoids. The method is rapid and reproducible. The utility of the method was demonstrated by its application to extracts from hypocotyls of young slash pine seedlings. [...]
