Abstract

Discovery of therapeutic or biotechnological agents is crucial in aiding the research of disease therapy. This thesis entails two projects, the first project focuses on utilizing a magnetic screening-based assay to identify peptoid-based compounds from a one-bead-one-compound (OBOC) library with specificity to human C-reactive protein, four initial hits were found, which was then repeated with a quantum dot assay that did not confirm binding of the hits to CRP due to technical concerns. The second project takes a peptoid-based hit GUCM-1 discovered from a previous screening assay with specificity for toll-interleukin 1 receptor (TIR) domain containing adaptor protein (TIRAP), an adaptor protein with a crucial role in the toll-like receptor 4 (TLR4) signal transduction pathway and studied it for its functional effects on a human myeloid cell line, results were inconclusive and further studies are needed to validate these findings.
