Idiopathic pulmonary fibrosis is a lung disease that affects the air sacs in the lungs and their interstitial. Symptoms of IPF include scarring of lung tissue, which impedes oxygen intake, and resultantly the body lacks oxygen. The disease’s pathogenesis is unknown, and resultantly there is no current cure. Typical lifespan after diagnosis is 3-5 years (nlhibi.nih.gov). My project aims to identify novel biomarkers for IPF, which is the first step towards designing better diagnosis options for patients. Current methods of accurate diagnosis rely on invasive methods such as biopsies to check histologically for scarring. Currently I am combing research databases for microarray studies that study the gene expression levels of tens of thousands of genes in human lungs with IPF versus controls. A meta-analysis is used to determine the best biomarkers for IPF across multiple studies. Highly upregulated genes on the plasma membrane of cells are potential biomarkers. In the lab, I will confirm the increased protein level of these upregulated genes using immunohistochemical staining, which involves using antibodies to target and highlight the presence of my target. After this is done, potential biomarkers will be identified, supported by a meta-analysis of several microarray studies and physical lab testing. Hopefully my efforts will aid in the therapeutic targeting of these biomarkers, which can be used in designing bedside, convenient, and early diagnosis options for patients with IPF.
