Abstract

Synthetic nucleosides have been used in this study to probe the active center of RNA polymerase. Results have been presented indicating: (1) the percent inhibition and the type inhibition of all inhibitors; (2) the drug-enzyme molar ratio for affinity labeling of the active center of RNA polymerase; (3) presumptive evidence that a cysteine residue is located in the active center. Additionally, atomic fluorescence has been used: (1) to confirm the presence of two (2) moles of Zn in every mole of enzyme; (2) to detect the removal of Zn from the active center and the substitution of Co+++, Mg++ and Zn++. The B' and B subunits were also shown to contain the Zn atoms.
