dc.description.abstractSynchronous budding cultures of Candida albicans were generated by releasing cells from stationary phase into fresh synthetic medium at 25°C. The cell cycle was monitored by measuring the DNA content of individual cells by flow microfluorometry. It was observed that cells synchronously formed germ tubes as a result of a temperature shift from 25°C to 37°C while in the G-l phase of the cell cycle. The capacity to form germ tubes was lost between 2 and 3 h when the cells entered the G-2 phase of the cell cycle. The cells were then committed to growth by budding. When cells returned to the G-l phase of the cell cycle the capacity to form germ tubes as monitored by a shift to 37°C returned. There was no loss in the capacity to form germ tubes after 2 h when DNA synthesis was inhibited by hydroxyurea or 5-fluorocytosine. [...]
