Abstract

ROCK1 is a kinase involved in regulating actin-myosin contraction, smooth muscle contraction, morphology, and cytoskeleton organization. Purpose: Optimization of expression will allow us to produce a homogenous Rock1 protein, which is absolutely required for protein crystallization as well as drug discovery studies in our lab. Conclusions. ROCK1 protein was only expressed in LONZA cell media. In LONZA, protein expression was best at 0.25 to 0.5 MOI. Protein expression is selective, suggesting conditions such as different cell media should be taken into account. Viral infection is best at a lower MOI so the virus does not kill the cells before the protein can be expressed.
