Abstract

Confocal laser scanning microscopy is a powerful and well-established technique with a wide range of applications. Structured illumination microscopy is a technique to extend the spatial resolution by a factor of two or potentially more beyond Abbe’s microscopy limit by using patterned illumination light. Certain aspects of each technique can be combined to give a new approach with advantageous features of each.

Theoretical work has been done to demonstrate the applicability of SIM principle to CLSM. This research focuses on experimental investigation and developing a state-of-the-art instrumentation that can be deployed to demonstrate this concept.

Research challenges are expected for all aspects of the proposed work. The anticipated ultimate impact of this research is a new feature that can be added to existing and future CLSM systems that can double the spatial resolution of these systems. Given the large number of existing CLSM for numerous applications, this feature may have a substantial commercial market.
